S1 is the interface between the eNodeB and the EPC. X2 is the interface between the eNodeB and the eNodeB.

Yes. If a cell needs to be set up, the S1 interface must be available so that the eNodeB can exchange signaling with the EPC. X2 is generally used in handover scenarios, but UE handovers can also be performed over the S1 interface.
